Jentadueto is a prescription medication that is classified as a combination product that contains a dipeptidyl peptidase 4 (DPP-4) inhibitor (linagliptin) and a biguanide (metformin). 

What is Jentadueto used for? 

It is used for the treatment of type 2 diabetes mellitus.

How does it work?

The Linagliptin component inhibits DPP4 which allows for increased levels of glucagon-like peptide-1 (GLP-1) and lower glucose levels in a glucose-dependent manner. Metformin increases glucose uptake and utilization to lower blood glucose levels.

Jentadueto is available as a brand only as either an immediate release or extended release tablet:

• IR: 2.5mg/500mg, 2.5mg/850mg, 2.5mg/1000mg tablets.

What is the recommended dosage?

What is the maximum dosage?

The maximum recommended dosage is 5mg linagliptin/2000mg metformin per day.

What if I miss a dose?

Take the medication as soon as possible if you have missed a dose. If it is almost time for your next dose, skip the missed dose and return back to your regular schedule. Never double the dose.

What should you know before you take Jentadueto?

Severe/disabling arthralgia has been reported with DPP-4 inhibitors. Onset can occur within a day to years after starting therapy.

Bullous pemphigoid development has been associated with DPP-4 inhibitors.

Vitamin B12 deficiency has been associated with long-term metformin use. Vitamin B12 serum concentrations should be periodically monitored with long-term therapy.

Acute pancreatitis cases have been reported with use. 

Caution in renal function. Dose adjustment/discontinuation of therapy may be needed.

• eGFR > 45 mL/minute/1.73m2: No dose adjustment needed. 
• eGFR 30 to 45 mL/minute/1.73m2:
  • Initiation: Not recommended.
  • Continuation: A metformin dose reduction of 50% is needed (maximum 1 g/day) and renal monitoring every 3 months is recommended. 
• eGFR < 30 mL/minute/1.73m2: Use is contraindicated. 

U.S. Boxed Warning: Metformin-induced lactic acidosis have been reported in postmarketing cases that have resulted in death. The risk is increased with renal dysfunction.

What are the side effects of Jentadueto?

Some common side effects include:

• Diarrhea
